Epidemiologic study on Candida species transmission from mother to their neonate in Lanzhou J Pract Stomatol 2009Vol.25(6)Nov,25

[Abstract] Objective: To investigate the candidal infection status in pregnant women in Lanzhou, and candidal transmission from mothers to their newborn infants. Methods: Vaginal fluid and saliva samples from 104 puerperas, as well as 104 saliva samples from their newborn infants, were collected. The candida species were cultured, isolated and identified using CHROMagar media. Further identification was taken using molecular biological technology. Result: In 81 of 312 specimens (104×2 from mothers and 104 from infants), candida species were found positively. 39.42%(41 cases)and 33.65%(35 cases) were observed in the vaginal and oral of puerperas respectively, and 21.15%(22 cases) in both vagina and oral cavity. The rate 4.81%(5 cases) was found in oral cavities of newborn infants. The distribution of candida species in positive specimens were 53 Candida albicans, 33 Candida glabrata, 2 Candida krusei and 1 Candida tropical. In 2 pairs of mother-infant, the same genotype of Candida ablicans was identified using PCR method. Conclusion: The Candida isolation rate of newborn infants and transmission rate from mother to their neonate in Lanzhou are higher than that in reported other areas. The colonization of candida in newborn infants is relevant to both horizontal and vertical transmission. It can decrease the likelihood of Candidal infection in newborn infants by controlling the Candidal transmission in hospital and preventing the infection in pregnant women.

Mutation analysis of the eda-A1 gene for hypohidrotic ectodermal dysplasia and construction of recombined eukaryotic expression vector West China Journal of Stomatology Vol.27 No.6 Dec.2009

[Abstract] Objective The purpose of this study was to clone and analyze mutation in the eda-A1 gene for hypohidrotic ectodermal dysplasia (HED), and to construct a new recombined eukaryotic expression vector (mutant M, wild W) as a basis for further study on the genetic function. Methods After total mRNA was extracted from peripheral blood lymphocytes from the HED affect patient and control, eda-A1 gene was amplified by reverse transcription polymerase chain reaction (RT-PCR) with a pair of specific primers containing the constriction enzyme sites of BamHⅠ and HindⅢ. When vector pcDNA3.1(-) and eda-A1 (M/W) were digested by BamHⅠ and HindⅢ respectively, eda-A1(M/W) fragment was then ligated to vector pcDNA3.1(-) and the new vector was named as pcDNA3.1(-)-eda-A1-M/W. Results eda-A1 gene was successfully cloned and a novel missence mutation (1091T→C) was identified, which changes the codon 306 from glutamine to proline; PCR, restrictive endonuclease analysis and  DNA sequencing were then performed to identify the recombinant eukaryotic expression vector pcDNA3.1(-)-EDA-A1-M/W, and the results were surely confirmed. Conclusion Our result indicates that the novel missense mutation in EDA is associated with the isolated tooth agenesis and provide preliminary explanation for the abnormal clinical phenotype at a molecular structural level. And also, the recombinant eukaryotic expression vector pcDNA3.1(-)-eda-A1-M/W was successfully constructed, which will be thereafter taken use of in further study on eda gene in odontogenesis.

Abstract

[Abstract] Objective: To study the antibiotic effect of the leaching solution of glycy- rrhizae and astragalus on one of the cariogenic bacteria -- Mutans streptococci and to compare to that of the oral bacteriostatic product from foreign country (MI). Methods: Glycyrrhizae and astragalus from Gansu was used as the raw material. Mutans streptococci were chosen as the experimental isolates that were cultured in the medium for 24 hours. The PH and OD600 values were then measured. Statis tical analysis was conducted using SPSS13.0. Result: The leaching solutions of glycyrrhizae and astragalus have inhibitory effects respectively on the growth and acid production of mutans streptococci, and have no significant differences com- paring that of foreign bacteriostatic product. Conclusion: The leaching solution of glycyrrhizae and astragalus respectively has antibacterial effect on Mutans strep- tococci.

Abstract

Objective: To compare the oral Candidas’presence and variance before and during treatment of fixed orthodontic appliances ( FOA) on teenagers.

Methods: The microorganism samples were taken before and after orthodontic treatment by culture of identification and molecular biology techniques through self2controlled method. Thus, the effects of orthodontic appliance on oral pathogenic yeasts were longitudinally studied.

Results: The percentage of people who carried some kinds of Candidas and the sum of colony forming unit was increased obviously after orthodontic treatment for 2 months. The sort of oral Candidas before and after treatment was also changed obviously.

Conclusion: Fixed orthodontic appliances can influence oral pathogenic yeasts in adolescence.

With the chang of medicinal pattern, research on psychological factors impacting diseases has become an important content in modern medicine. Oral health impact profile (OHIP) is subjected to the comprehensive assessment of oral diseases in the areas of physical, psychological and social function. It is well complementary for modern dentistry researches, and also leads to the much concerns on oral health related quality in the world. To explore the possibility of OHIP application in China, this paper aimed to describe the formation, development, features of OHIP, and to express the prospects in the near future of OHIP in Chinese.

Abstract

Objective: To study the cariogenic characteristics of Mutans Streptococci (MS) and Lactobacilli (LB) during the cariogenic development of the elderly. Methods: 136 dental carious samples were collected for isolation, cultivation, identification. CFU count was then conducted in the sorts by age, carious degree and carious activity. The mean count of MS and LB’s CFU, and the samples’ amount resulted from the questioned bacterium (bacteria) were statistically analyzed by SPSS 13.0 after logarithmic conversion to data. Results: 1, There was a significant difference in mean counts of SM and LB in the group of age, carious degree and carious activity, respectively (P<0.05). 2, There was a significant difference in the amount of commonly cariogenic samples by SM and LB in the elderly group (P<0.05), which was closely correlated with the carious degree (P<0.05). Conclusion: MS and LB are mutualism in the cariogenic development of the elderly, the median caries, and the serious caries respectively.

Abstract

Hypohidrosis ectodermal dysplasia (HED) is a rare congenital disorder that results in abnormalities in the structures of ectodermal origin tissues, such as teeth, eccrine sweat glands, hair and nails. However, most of studys in HED is presented in the form of clinic case reports. This article reviews the research progress of HED in types, genetic locations, proteins, and signaling pathways in odontogenesis.
